中文摘要:用RAPD技术对中国蔗蝗属3种蝗虫基因组DNA的多态性进行研究。在事先优化的反应条件下用12个随机引物扩增,共得到179条清晰稳定的多态性片段,片段长度为200—2000bp。统计这些片段,根据扩增片段的共享度计算出相对遗传距离指数,然后用NJ和UPGMA聚类方法对其进行分析,构建系统树,以确定这3种间的亲缘关系。结果表明:等歧蔗蝗与异歧蔗蝗二者的亲缘关系较近,而与斑角蔗蝗的关系较远。

Analysis of the Genetic Variations and Phylogenetic Relationships Among Three Species of Grasshopper Genus Hieroglyphus (Orthoptera: Acridoidea:Catantop idae) Using RAPD Markers

Abstract:Random amplified polymorphic DNA (RAPD) techniques were applied to assess geneti c variations among three species in the grasshopper genus Hieroglyphus Kraus s, 1877. Amplifications with 12 primers under predetermined optional reaction co nditions gave 179 RAPD fragments ranging from 200 to 2000 bp in length. The pr oportion of RAPD fragments shared (F) and the genetic distance (D) among three species were calculated. Phylogenetic trees based on the genetic distance matric es with the method of NJ and UPGMA were constructed in order to determine the ph ylogenetic relationships among three species of this genus. The results indicate d that the phylogenetic relationship between Hieroglyphus banian (Fabricius )and H. tonkinensis I. Bolivar was more closer than that with H. annulico rnus (Shiraki), but was far to H. annulicornus .
